Binding studies with 45Ca and exchange measurements with 35S-labelled Ca2+ and Ca2+ have shown that the Ca2+ storage pool of bovine cardiac sarcoplasmic reticulum (SR) possesses some of the properties expected for the sarcoplasmic reticulum (SR) Ca2+-ATPase. An equilibrium (bound:total Ca2+ ratio of 4:1) was reached very rapidly after addition of Ca2+ to SR vesicles suspended in buffers with 0.2 to 0.5 mM Ca2+. This ratio was independent of the Ca2+ concentration, the concentration of SR vesicles and the minimum free Ca2+ concentration during the exchange experiment. The 45Ca exchange rate was not affected by Ca2+ concentrations below 0.5 mM and was more than twice as fast as the exchange rate in vesicles stored at 0.1 mM free Ca2+. Exchange was inhibited by 0.3 mM EGTA, and SR-dependent ATP hydrolysis stimulated exchange in EGTA-treated SR vesicles.
