Polysialic acid (PSA), a homopolysaccharide of sialic acids, is an interesting glycoprotein modification produced by the bacteria Lactobacillus fermentum subsp. frumentarius. L. fermentum subsp. frumentarius has been the source of PSA for several years. Therefore, the optimization of the production of PSA is essential to guarantee a continuous supply for its implementation in the pharmaceutical industry. This study presents the optimization of the PSA production process using a fed-batch strategy in a 1 L-scale fermenter. The optimization study was guided by response surface methodology (RSM). Optimization of cell growth was evaluated by cell concentration and PSA yield after 72 hours, and by PSA production rate in the first 48 hours. For PSA optimization, 20 strains were evaluated and only seven strains were able to produce PSA. The maximum PSA production (78.83+/-1.93 mg/L) was obtained at pH 7.4, glucose (1.0%) and nitrogen (0.75%) concentration in the fermentation medium, respectively. The three-dimensional matrix plot showed that the most suitable design for PSA production included glucose, nitrogen and pH. The results showed that the PSA production rate is determined by the nitrogen concentration in the fermentation medium, which means that the optimization of PSA production has priority over the optimization of nitrogen concentration in the medium. The fed-batch strategy used in this study was successful in producing PSA with good reproducibility, and the optimal conditions obtained are promising for the continuous production of PSA and its possible use in the production of PSA-based pharmaceuticals.Here’s what it looks like when the best restaurant breaks
